آنزیمهای برشی یا آندونوکلئاز محدودکننده اغلب از گروه هیدرولازهاست که بهطور اختصاصی، توالی خاصی از DNA را شناسایی کرده و آن را برش میدهند.
در صورتی که توالیهایی توسط عواملی مثل متیلهشدن تغییر یافته باشند مورد شناسائی آنزیم قرار نمیگیرند.
آنزیمهای محدودکننده بهطور متداول به ۴ گروه دستهبندی میشوند. این ۴ گروه از نظر ساختار، جایگاه شناسایی و چگونگی برش با یکدیگر متفاوتند. به منظور ایجاد برش در DNA، آنزیمهای محدودکننده باید ستون قند-فسفات (sugar-phosphate backbone) را در هر دو رشته قطع کنند.
این آنزیمها در باکتریها و آرکیها یافت شده و یک مکانیسم دفاعی علیه ویروسهای مهاجم ارائه میدهند، در یک پروکاریوت، آنزیمهای محدودکننده به صورت انتخابی سبب برش DNA خارجی در فرایندی که restriction نامیده شدهاست، میشوند. در حالیکه DNA میزبان توسط آنزیم اصلاحکننده (یک متیل ترنسفراز) که سبب تصحیح DNA پروکاریوت میشود، حفظ میشود. این دو فرایند سبب شکلگیری سیستم مدیفیکاسیون محدودکننده (restriction modification system) میشوند

Recommended Protocol for Digestion
Add:
nuclease-free water 16 μL
10X Buffer Tango 2 μL
DNA (0.5-1 μg/μL) 1 μL
BspLI 0.5-2 μL
Mix gently and spin down for a few seconds.
Incubate at 37°C for 1-16 hours.
The digestion reaction may be scaled either up or down.
Recommended Protocol for Digestion of PCR Products
Directly after Amplification
Add:
PCR reaction mixture 10 μL (~0.1-0.5 μg of DNA)
nuclease-free water 18 μL
10X Buffer Tango 2 μL
BspLI 1-2 μL
Mix gently and spin down for a few seconds.
Incubate at 37°C for 1-16 hours.
